The presynaptic terminal, often referred to as the axonal terminal, releases neurotransmitters into the synaptic cleft in order to deliver signals to the postsynaptic neuron. In the cytoplasm of the presynaptic membrane, there are many synaptic vesicles, some microfilaments and microtubules, mitochondria and slippery endoplasmic reticulum, etc.Synaptic vesicles are characteristic structures at the front of the synapse that contain chemicals called neurotransmitters.
Synaptosomal-associated protein-25 (SNAP25) is a presynaptic protein that takes part in vesicular exocytosis, neurite outgrowth, and long-term potentiation.

Synapsins are neuron-specific phosphoproteins that are exclusively associated with small synaptic vesicles. Synapsin 1 is one of the most specific markers of synapses throughout the central and peripheral nervous system. In addition to synaptic nerve terminals, the protein is also present in certain sensory nerve endings. Synapsin 1 interacts with vesicle membranes as well as with actin and spectrin. Because of Synapsin 1's characteristic sub-cellular localization, antibodies against this protein have been used as one of the most reliable markers for pre-synaptic vesicles.

Synaptotagmin 1 is a 47,442 dalton protein (422 amino acids) associated with pre-synaptic terminals of the PNS and CNS. Synaptotagmin-1 binds calcium and initiates the fusion of vesicles with the pre-synaptic membrane. This protein also is believed to be the target of proteolytic activity associated with Botulinum neurotoxin A.
