Introduction to Superoxide Dismutase (SOD1)

Amyotrophic lateral sclerosis (ALS), also known as Lou Gehrig's disease, is a progressive, fatal neurodegenerative disease marked with the loss of upper and lower motor neurons. The superoxide dismutase (SOD1) gene, the major antioxidant gene, was the first gene to be associated with a familial form of ALS. Other genes associated with inherited ALS include TDP-43 and FUS. Numerous experiments have shown that full-length apo SOD1 can be converted into amyloid fibrils by incubation with reducing agents in vitro. Importantly, SOD1 protofibrils formed in vitro can incorporate into cells and spread between cells. Furthermore, SOD1 protofibrils produced under reducing conditions share the same pathological properties as ALS inclusion bodies, such as the ability to induce mitochondrial damage, cause neuroinflammation and activate microglia to trigger ALS neurodegeneration. Therefore, the structural determination of SOD1 protofibrils is important for understanding the pathogenic mechanisms of SOD1 in familial and sporadic ALS.

Advantages of Our SOD Pre-Formed Fibrils (PFF)

  • We prepared homogeneous cytotoxic amyloid protofibrils from full-length recombinant apo-human SOD1 under reduced conditions and determined the atomic structure using cryo-electron microscopy. Our product can be used to study the SOD1 transition between the physiological and protofibril states.
  • We have determined the atomic structure of cytotoxic SOD1 amyloid protofibrils by cryo-electron microscopy, allowing you to better understand the product structure.
  • Our SOD1 preformed protofibrils (PFF) products provide you with the material to study SOD1 monomer aggregation.
