Oligodendrocytes are a type of neuroglia whose main functions are to provide support and insulation to axons in the central nervous system of some vertebrates. Oligodendrocytes arise during development from oligodendrocyte precursor cells (OPCs), which can be identified by their expression of a number of antigens, including the ganglioside GD3, the NG2 chondroitin sulfate proteoglycan, and the platelet-derived growth factor-alpha receptor subunit (PDGF-alphaR). Mature oligodendrocytes are broadly classified into either myelinating or non-myelinating satellite oligodendrocytes. Precursors and both mature types are typically identified by their expression of the transcription factor OLIG2.
Oligodendrocyte transcription factor (OLIG2) is a basic helix-loop-helix (bHLH) transcription factor. OLIG2 is mostly expressed in restricted domains of the brain and spinal cord ventricular zone which give rise to oligodendrocytes and specific types of neurons.

A2B5 is a cell surface ganglioside epitope expressed in developing thymic epithelial cells, oligodendrocyte progenitors, and neuroendocrine cells. It is an excellent marker for thymic epithelial cells, oligodendrocyte progenitors and neuroendocrine cells.

Myelin basic protein (MBP) is a major myelin constituent produced by oligodendrocytes in the central nervous system (CNS). Expression of MBP was considered to be a marker for oligodendrocyte differentiation and myelination in the developing CNS and it has long been considered as a marker of active demyelination.

Sox proteins are characterized by possession of a DNA-binding domain with similarity to the high-mobility group domain of the sex determining factor SRY. During development Sox10 first appeared in the forming neural crest and continued to be expressed as these cells contributed to the forming PNS and finally differentiated into Schwann cells. Adenomatous Polyposis Coli (APC) is a +TIP protein that promotes microtubule assembly and has been studied extensively for its roles in biology and disease. CNPase is found in high levels in the brain and peripheral nerve, almost exclusively in oligodendrocytes and Schwann cells. CNPase is the third most abundant protein found in the central nervous system myelin. It is found early in oligodendrocyte development and continues to be found in adult animals.

Nogo-A is expressed in oligodendrocytes, in the inner and outer loops of myelin sheaths, and in some neuronal populations such as dorsal root ganglion cells, retinal ganglion cells, and ventral horn neurons in spinal cord. Nogo-A was originally identified because of its ability to prevent axonal regrowth and sprouting. Treatment with antibodies recognizing the N-terminal epitope of Nogo-A (IN-1) resulted in improved regenerative fiber outgrowth in rats with spinal cord trauma.

Myelin proteolipid protein (PLP) is identified as a specific and largely heat-resistant marker for detection of food contaminations by CNS tissue. PLP is a component of oligodendritic glial sheaths of neuronal processes that is specifically expressed in the CNS.

Oligodendrocyte-myelin glycoprotein (OMgp) is expressed on the surface of oligodendrocytes and neurones and is thought to inhibit axonal regeneration after brain injury in adult, like Nogo and myelin-associated glycoprotein (MAG). Previous study suggests that OMgp is a late marker of myelination, which could be implicated in the arrest of oligodendrocyte proliferation, arrest of myelination or compaction of myelin.
