Gamma-synuclein is a protein encoded in humans by the SNCG gene and is one of three members of the synuclein family (α-synuclein/SNCA, β-synuclein/SNCB, and SNCG). Gamma-synuclein is mainly found in the peripheral nervous system (primary sensory, sympathetic, and motor neurons) and in the retina, and is commonly expressed in breast tumors. It is also found in the brain, ovarian tumors, and olfactory epithelial cells. γ-Synuclein is also a marker for tumorigenesis. It has been implicated in neurodegenerative disease pathogenesis. Changes in the expression of γ-synuclein have been observed in the retina of patients with Alzheimer's disease. γ-Synuclein is thought to exist in a naturally unfolded conformation and readily aggregates in solution to form small soluble oligomers. These inclusions are present in neurons and synapses in the amygdala and substantia nigra of the human brain that contain Lewy bodies. Accumulation of gamma-synuclein is also seen in some cases of ALS, where it can accumulate in inclusion bodies and lead to motor neuron death. α- and γ-synuclein are thought to interact and promote aggregation of each other.
