The growth cone is a highly dynamic, actin-rich structure capable of recognizing and responding to a variety of guidance cues. It consists of two major domains, the peripheral and central domains, which are characterized by specific cytoskeletal components. The peripheral domain is rich in actin filaments, while the central domain mainly contains microtubules, mitochondria, and various other organelles. Neuronal growth cones execute important steps in neural wiring, including axonal growth and pathfinding, and accurate synaptogenesis.
Growth-associated protein (GAP-43) is a proven marker for developing and regenerating neurons, and to a lesser extent, reactive glial cells. It is expressed in conditions of embryonic growth, during axonal regeneration, and even at maturity in certain areas of the brain known to exhibit synaptic plasticity and is used widely to specifically label injured neurons and to score neuronal regeneration.
