Microglia are cells of mesodermal/mesenchymal origin that migrate into the CNS to become resident macrophages within the unique brain microenvironment. Microglia are highly dynamic cells that interact with neurons and nonneuronal cells.
Microglia express a wide array of receptors and thus respond to pleiotropic stimuli ranging from neurotransmitters to cytokines and plasma proteins. They play a crucial role in the healthy brain as regulators of synaptic functions and phagocytosis of newborn neurons, with important implications in synaptic plasticity and adult neurogenesis. In disease, they play a crucial role in neurodegenerative and neuroinflammatory conditions. Their interactions with T cells are a major component of the development of brain autoimmunity, while their pathogenic interactions with neurons via induction of ROS and iNOS play a crucial role in neurodegeneration.
Ionized calcium-binding adapter molecule 1 (Iba1) is a 147-amino-acid calcium-binding protein widely in use as a marker for microglia. It has actin-crosslinking activity and is involved in aspects of motility-associated rearrangement of the actin cytoskeleton.

CD11b is an integrin family member which pairs with CD18 to form the CR3 heterodimer. CD11b is expressed on the surface of many leukocytes including monocytes, neutrophils, natural killer cells, granulocytes and macrophages, as well as on 8% of spleen cells and 44% of bone marrow cells. It is an excellent marker of macrophages and microglia.

CD115 (macrophage colony-stimulating factor receptor, or CSF-1 receptor) is the sole cell-surface receptor identified for CSF-1, the predominant growth factor regulating the survival, growth and differentiation of myeloid lineage cells comprising monocytes, macrophages, DCs and osteoclasts. Purinergic Receptor P2Y12 (P2RY12), a P2 purinergic receptor, is introduced as a specific marker for microglial cells under normal and pathologic conditions. P2RY12 belongs to the family of P2 purinergic receptors, consisting of seven transmembrane G protein coupled receptors (GPCRs) that contribute to ATP-and ADP-mediated cell migration in vitro. P2RY12 is expressed in activated platelets and notoriously, in microglial cells.

Sialic acid-binding immunoglobulin-like lectin H (Siglec-H) is expressed by parenchymal microglia from developmental stages to adulthood, and the expression was maintained in activated microglia under injury or inflammatory condition. Siglec-H expression was absent from CNS-associated macrophages and CNS-infiltrating monocytes, except for a minor subset of cells. It is a microglia-specific marker that discriminates microglia from CNS-associated macrophages and CNS-infiltrating monocytes.

TREM2 is a receptor glycoprotein that belongs to the immunoglobulin superfamily. In the brain, TREM2 is expressed exclusively by myeloid cells, including microglia and macrophages.
