Schwann cells (SCs), which surround nerve fibers in the normal peripheral nerve tissue, prO4vide structure support, conduct nervous impulses along axons, clear debris after peripheral nerve axon damage, and guide axonal regeneration. Previous studies have reported several SC-specific markers, and the most commonly used are S100, MBP, MPZ, P75NTR, NCAM, GAP43, Sox10, Oct6, Krox20 and Sox2. The SC specific markers in various SC stages are different but partially overlapping.

Growth-associated protein (GAP-43) is a proven marker for developing and regenerating neurons, and to a lesser extent, reactive glial cells. It is expressed in conditions of embryonic growth, during axonal regeneration, and even at maturity in certain areas of the brain known to exhibit synaptic plasticity and is used widely to specifically label injured neurons and to score neuronal regeneration.

Oct6 is present in cytoplasm of Schwann cells associated with normal-appearing myelinated nerve fibers, but not in nuclei. Expression is seen in nuclei of Schwann cells in the early phase of acute axonal degeneration; nuclear expression peaks at the regenerative stage.

Myelin basic protein (MBP) is a major myelin constituent produced by oligodendrocytes in the central nervous system (CNS). Expression of MBP was considered to be a marker for oligodendrocyte differentiation and myelination in the developing CNS and it has long been considered as a marker of active demyelination.

Myelin protein zero (P0, MPZ) is a single membrane glycoprotein, which is a major structural component of the myelin sheath in the peripheral nervous system (PNS). Myelin protein zero is expressed by Schwann cells and accounts for over 50% of all proteins in the peripheral nervous system, making it the most common protein expressed in the PNS.

NCAM is a differentiation marker of postmigratory immature neurons in the developing human nervous system. NCAM was expressed by non-myelinating Schwann cells from normal nerves and over-expressed by Schwann cells from patients with chronic axonal neuropathies and Schwannomas.

Sox proteins are characterized by possession of a DNA-binding domain with similarity to the high-mobility group domain of the sex determining factor SRY. During development Sox10 first appeared in the forming neural crest and continued to be expressed as these cells contributed to the forming PNS and finally differentiated into Schwann cells. In the CNS, Sox10 transcripts were originally confined to glial precursors and later detected in oligodendrocytes of the adult brain.

S100 calcium-binding protein B (S100B) is a protein of the S-100 protein family. Nerve growth factor receptor p75NTR is a major neurotrophin receptor involved in manifold and pleiotropic functions in the developing and adult central nervous system (CNS). Depending on the complex interactions with other receptors and on the cellular context, p75NTR is capable of performing contradictory tasks such as mediating cell death as well as cell survival. In parallel, as a prototype marker for certain differentiation stages of Schwann cells and related CNS aldynoglial cells, p75NTR has recently gained increasing notice as a marker for cells with proposed regenerative potential in CNS diseases, such as demyelinating disease and traumatic CNS injury.
