CRMP1 Introduction
Collapsin response mediator protein 1, encoded by the CRMP1 gene, is a human protein of the CRMP family.This gene encodes a member of the cytoplasmic phosphoprotein family that is only expressed in the nervous system. The encoded proteins are considered part of the signal transduction pathway and are associated with the collapse of the growth cone during signal induced neural development. Alternative splicing leads to multiple transcriptional variants. CRMP1 mediates reelin signaling in cortical neuron migration. CRMP1 deficient mice exhibit long-term potentiation impairment and spatial learning and memory impairment. Collagenase reaction mediator protein 1, abbreviated as CRMP1, is an important member of the CRMP protein family (including CRMP1-5). It is a cytoplasmic phosphoprotein highly expressed in the nervous system. In the brains of AD patients, CRMP1 can be hyperphosphorylated and abnormally aggregated. This abnormal modification may impair its normal function, leading to impaired intracellular transport and synaptic degeneration, thereby exacerbating tau pathology and neuronal dysfunction.

CRMP1 typically exists in the form of homo - or hetero tetramers (usually forming complexes with other family members such as CRMP2). Each monomer contains an N-terminal domain and a C-terminal domain, which allows it to interact with various partner molecules.

CRMP1 is a key intracellular mediator in the Semahorin 3A (Sema3A) signaling pathway. Its core function can be summarized as: converting extracellular guiding signals (Sema3A) into intracellular instructions for cytoskeleton rearrangement.
Specific Mechanisms of Action
Signal reception and transduction: When Sema3A binds to receptor complexes on the surface of neurons (such as Neuropilin-1/plexin-A), it activates a series of intracellular signaling events.
Regulating microtubule dynamics: CRMP1 is recruited near signal complexes and regulated through mechanisms such as phosphorylation. Activated CRMP1 can directly bind to microtubule protein dimers, promoting microtubule assembly and stability, thereby determining the direction of axonal growth.
Connecting vesicle transport: CRMP1 also interacts with motor proteins such as Kinesin to participate in axonal specific vesicle transport, providing new membrane materials for the advancement of growth cones.
Integration with other signals: The function of CRMP1 is finely regulated by phosphorylation of various kinases (such as GSK-3 β, Cdk5, ROCK), making it a hub for integrating multiple axonal guidance signals (such as Netrin, BDNF).
