Understanding the magnitude, breadth, and functionality of an antigen-specific immune response is central to the development of vaccines, immunotherapies, and cell and gene therapy products. Creative Diagnostics' ELISpot and FluoroSpot laboratory provide sponsors with single-cell resolution immune monitoring data to support discovery research, preclinical studies, and clinical trial immunogenicity endpoints.
We work as an extension of your project team -- from assay feasibility and antigen/peptide panel design through sample logistics, testing, and final data delivery -- applying rigorous, SOP-driven laboratory practices at every step.
The Enzyme-Linked ImmunoSpot (ELISpot) assay is a well-established immunological method that enables direct visualization and enumeration of individual cells secreting a specific analyte -- most commonly a cytokine such as IFN-γ, IL-2, IL-4, IL-5, or Granzyme B -- in response to antigenic or mitogenic stimulation. Because each spot on the membrane corresponds to a single responding cell, ELISpot provides both the frequency and functional profile of antigen-specific lymphocyte populations within a sample, rather than an averaged, bulk-culture readout.
FluoroSpot extends the same capture principle using fluorescently labeled detection reagents, allowing two or more analytes to be measured simultaneously within the same well. This makes it possible to identify multi-functional or polyfunctional responding cells -- for example, cells that co-secrete IFN-γ and IL-2 -- from a single sample well.
Fig.1 Simplified schematic of the ELISpot assay principle: Antigen-specific cells secrete cytokine that is captured locally on an antibody-coated membrane, forming a visible spot.

T cell response profiling against vaccine antigens or peptide pools, longitudinal immune monitoring across clinical study visits, and screening of candidate antigens during preclinical development.
Detection of tumor antigen- and neoantigen-specific T cell responses, and pharmacodynamic monitoring of T cell reactivation following checkpoint inhibitor or combination therapy.
Functional potency characterization of CAR-T and TCR-engineered cell products, and assessment of anti-vector or anti-transgene cellular immune responses following AAV or other viral vector administration.
Pathogen-specific T cell response characterization for infectious disease research, and evaluation of autoantigen-reactive cell populations in autoimmune disease studies.

Every study begins with a scientific consultation to align on antigen selection, cytokine panel, sample type, and study timelines. Samples are processed under standardized operating procedures, with quality control checkpoints at cell isolation, plating, and post-development imaging stages, ensuring consistent, well-documented data from receipt through final report.
