CDIA™ Recombinant Adenovirus (rAdv) Rapid Test Kit is a rapid chromatographic immunoassay for qualitative detection of rAdv and rapid estimation of rAdv titers. The kit can recognize all 3, 5, 7, 11, 40 and 41 subtypes of adenovirus.
Recombinant adenoviruses have been used to infect many mammalian cell types (both replicative and non-replicative) for high expression of the recombinant protein. Recombinant adenoviruses are especially useful for gene transfer and protein expression in cell lines that have low transfection efficiency with liposome. Titration of Adenovirus stocks is important for maintaining consistency between experimental samples and achieving the correct level of transient expression. Also, when producing viral stocks it is important to know the titer of infectious particles for sucessful virus production. Traditionally, infectivity particles are measured in culture by a plaque-forming unit assay (PFU) that scores the number of viral plaques as a function of dilution. These methods are time-consuming (10 days), require a long infection period, and suffer from a high degree of inter-assay variability and are affected by virus-cell interactions. CDIA™ Recombinant Adenovirus (rAdv) Titration Rapid Test Kit is designed around a hexon-specific antibody, which is used to label infected cells. Hexon protein is encoded by the Adenovirus genome and is an essential component of the Adenovirus capsid required for Adenovirus replication.
Principles of Testing
CDIA™ Recombinant Adenovirus (rAdv) Rapid Test Kit is a rapid chromatographic immunoassay for qualitative detection of rAdv and rapid estimation of rAdv titers. The membrane is pre-coated with anti-Hexon antibodies on the test line region, anti-mouse secondary antibodies. During testing, specimen reacts with the particles coated with anti-Hexon antibodies. The mixture migrates upward on the membrane by capillary action to react with anti-Hexon antibodies on the membrane and generate a colored line. If the specimen contains Hexon protein, the result is visible as red line within ~10 minutes in the test line on the membrane. The solution continues to migrate to encounter a control reagent that binds a control conjugate, thereby producing another red control line. Two color bands indicate the presence of rAdv.
