Introduction
ClpC is an AAA+ ATPase chaperone in Staphylococcus aureus that forms hexameric complexes with protease subunits (e.g., ClpP). It uses ATP hydrolysis to unfold and translocate damaged/misfolded proteins into ClpP for degradation, maintaining proteostasis during stress (e.g., heat, antibiotics). ClpC also regulates virulence factors (e.g., toxin secretion) and activates key adaptor proteins like MecA for β-lactam resistance . Essential for bacterial survival, it represents a target for cyclomarin-type antibiotics that disrupt proteolysis, sensitizing MRSA to host defenses.
