Introduction
Replication initiation protein (RepD) is a 53.5 kDa plasmid-encoded protein (311 amino acids) in Staphylococcus aureus that functions as a specific topoisomerase to initiate plasmid replication. It binds plasmid DNA origin sequences and catalyzes strand cleavage/religation, enabling replication loop formation. RepD is rate-limiting for plasmid propagation in vivo and essential for mobilizing antibiotic resistance genes. Recombinant RepD (full-length, N-terminal 6×His-SUMO-tagged) is expressed in E. coli with >90% purity for studying plasmid dynamics and horizontal gene transfer mechanisms.
