Introduction
Foldase protein PrsA is a membrane-anchored lipoprotein in Staphylococcus aureus that acts as a chaperone for post-translocational protein folding. It stabilizes and facilitates the secretion of virulence factors, notably protein A (SpA), by directly interacting with its N- and C-terminal domains. Deletion of prsA reduces SpA secretion, stability, and immunoglobulin-binding capacity, impairing immune evasion. PrsA also dimerizes via these domains, enhancing its function. Recombinant PrsA is expressed in E. coli, yeast, and baculovirus systems for mechanistic studies. Targeting PrsA disrupts virulence, offering therapeutic potential against antibiotic-resistant infections.
