General Description
RNP-70 antibody (anti-U1-RNP-70 kD) is an autoantibody directed against the 70-kD protein subunit of the U1 small nuclear ribonucleoprotein (U1-nRNP) complex, which is essential for pre-mRNA splicing.
Clinical significance
• Hallmark of mixed connective-tissue disease (MCTD / Sharp syndrome) – found in 95–100 % of patients with high titers.
• Also seen in systemic lupus erythematosus (~30–50 %), systemic sclerosis (~25–30 %), and polymyositis/dermatomyositis (~10–20 %).
• High-titer anti-RNP-70 strongly supports the diagnosis of MCTD and helps differentiate it from pure SLE or SSc.
• Low-titer positivity indicates autoimmune activity but requires correlation with clinical findings and additional autoantibody profiles.
