Applications
Application Notes
Suitable for use in PK bridging ELISA.
We recommend the following for sandwich ELISA (Capture - Detection):
CABT-ZB242 - CABT-ZB243; CABT-ZB242 - CABT-ZB244
Recommended Working Concentration: 0.01-0.1 μg/ml
General Notes
We recommend the following antibody pair to develop a pharmacokinetic (PK) bridging assay to measure free drug: CABT-ZB242 - CABT-ZB243; CABT-ZB242 - CABT-ZB244
CABT-ZB242 is recommended for use as a capture antibody when paired with CABT-ZB243 or CABT-ZB244 as a detection antibody.
Rituximab (Rituxan) is a humanized monoclonal antibody that is approved by the U.S. Food and Drug Administration for the treatment of hematological cancers and autoimmune diseases. When Rituximab binds to CD20 on the surface of normal and malignant B cells, natural killer (NK) cells is activated to destroy these B cells. It is therefore used to treat diseases with too many B cells, overactive B cells, or dysfunctional B cells.

ELISA, PK study
CABT-ZB242 binds with Rituximab. While the antibody does not recognize the human lgG Fc fragment (data not shown).
Coating antigen: Rituximab, 1 μg/ml.
CABT-ZB242 dilution start from 2,000 ng/ml, EC50=7.819 ng/ml.
ELISA, PK study
Standard curve of Rituximab Sandwich ELISA. The Rituximab Sandwich ELISA assay is developed by using CABT-ZB242 and CABT-ZB243 as capture and detection antibodies, respectively. In this ELISA assay, CABT-ZB243 was labeled with Biotin.
The sensitivity of detecting Rituximab less than 1 ng/ml.
