THE EFFECTS OF GNRH AND CARAZOLOL ON OVULATION AND PREGNANCY RATES IN COWS

Abstract
The effects of the intramuscular injection of the gonadotropin-releasing hormone (GnRH) analogue ''Buserelin'' and the intravenous administration of carazolol, 30 minutes before insemination, on the ovulation and pregnancy rates were investigated. The study involved 335 cows which were clinically normal at the time of artificial ir?semination. Control animals received physiological saline. Milk samples were taken on days 0, 3, 5 and 7 of the oestrus cycle and examined in a progesterone assay. Of the 133 animals treated with carazolol, 14.2% exhibited delayed ovulation as determined by rectal examination. Of the 85 animals treated with GnRH 16.5% exhibited delayed ovulation compared with 18.8% in the control group. A strong correlation was found between the results of rectal examination and hormone assays in determining the results of ovulation. Of the 85 animals treated with GnRH 70.5% were diagnosed as pregnant by rectal examination compared with 70.7% in the carazolol treated group and 61.5% in the control group. These differences were not statistically significant, but indicate a strong positive effect of the treatments. In animals with delayed ovulation the conception rate in the carazolol treated animals was 63.2% compared with 64.3% in rite GnRH group and 45.4% in the control animals.
Simultaneous detection of residues of beta-adrenergic receptor blockers and sedatives in animal tissues by high-performance liquid chromatography/tandem mass spectrometry

Abstract
A method using high-performance liquid chromatography coupled with tandem mass spectrometry (HPLC-MS/MS) was developed to screen and confirm residues of multi-class veterinary drugs in animal tissues (porcine kidney, liver, muscle; bovine muscle). Thirty target drugs (19 beta-blockers, 11 sedatives) were determined simultaneously in a single run. Homogenized tissue samples were extracted with acetonitrile and purified using a NH2 solid-phase extraction cartridge. An Acquity UPLC (TM) BEH C18 column was used to separate the analytes. followed by tandem mass spectrometry using an electrospray ionization source in positive mode. Recovery studies were done at three fortification levels. Overall average recoveries in pig muscle, kidney, and liver fortified at three levels from 76.4% to 118.6% based on matrix-fortified calibration with coefficients of variation from 2.2% to 19.9% (n = 6). The limit of quantification of these compounds in different matrices was 0.5-2.0 mu g/kg. This method was successfully applied in screening and confirming target drugs in >200 samples.
