Intended Use

The Human PSA-total (Prostate Specific Antigen, total) ELISA kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of human PSA-total in serum, plasma, and cell culture supernatants.

Contents of Kit

1. Human PSA-total Microplate, 1 Plate. 96 wells (12 x 8) coated with anti-Human PSA-total. Storage/Stability after preparation: 1 month at 4°C. Note: Return unused wells to the pouch containing desiccant pack, reseal along entire edge.
2. Human PSA-total Standard Protein, 2 vials. Each vial is enough to run each standard in duplicate. Storage/Stability after preparation: 1 week at -80°C.
3. Human PSA-total Detection Antibody, 2 vials. Each vial is enough to assay half the microplate. Storage/Stability after preparation: 5 days at 4°C.
4. Wash Buffer (20X), 25 mL. 20X concentrated solution. Storage/Stability after preparation: 1 month at 4°C.
5. HRP-Streptavidin, 200 μL. 500X concentrated HRP-conjugated streptavidin. Storage/Stability after preparation: Do not store and reuse.
6. TMB One-Step Substrate Reagent, 12 mL. 3,3,5,5'-tetramethylbenzidine in buffer solution. Storage/Stability after preparation: N/A.
7. Stop Solution, 8 mL. 0.2 M sulfuric acid. Storage/Stability after preparation: N/A.
8. Assay Diluent A, 30 mL. 1X diluent buffer. Storage/Stability after preparation: N/A.
9. Assay Diluent B (5X), 15 mL. 5X concentrated diluent buffer. Storage/Stability after preparation: 1 month at 4°C.
10. 96-Well Plate Sealing Film, 2 films. Microplate adhesive sealing film. Optional use for overnight incubations. Storage/Stability after preparation: N/A.

Storage

The entire kit may be stored at -20°C for up to 1 year from the date of shipment. Avoid repeated freeze-thaw cycles. The kit may be stored at 4°C for up to 6 months. For extended storage, it is recommended to store at -80°C. For prepared reagent storage, see Reagents And Materials Provided.

Sensitivity

The minimum detectable dose of Human PSA-total was determined to be 8 pg/ml. Minimum detectable dose is defined as the analyte concentration resulting in an absorbance that is 2 standard deviations higher than that of the blank (diluent buffer).

Standard Curve

These standard curves are for demonstration only. A standard curve must be run with each assay. Standard curves were generated in Assay Diluent A and Assay Diluent B respectively (see standard curve figures).
