Phosphatidylinositol (PI) is a membrane phospholipid involved in regulating key cellular processes. Its inositol head can be uniquely phosphorylated at the 3, 4, and 5 hydroxyl residues, forming seven different phosphoinositides. Phosphatidylinositol (3,4,5)-trisphosphate (PIP3) is one of them. PIP3 is generated from phosphatidylinositol-4,5-bisphosphate (PIP2) by the action of phosphatidylinositol 3-kinase (PI3K). PIP3 can also be dephosphorylated back to PIP2 by phosphatidylinositol-3,4,5-trisphosphate 3-phosphatase (PTEN), thus terminating signal transduction.
PIP3 plays a critical role in cell signal transduction, particularly in pathways involving cell growth, differentiation, metabolism, survival, and movement. As a second messenger, PIP3 regulates various physiological processes by binding to signaling proteins such as Akt and PDK1 on the cytoplasmic membrane, thereby activating a series of downstream signaling pathways. PIP3 binds to Akt kinase, and upon activation, Akt can inhibit apoptosis and promote cell growth and division. PIP3 can also recruit PDK1 to the cell membrane and, by binding to it, promote PDK1 activation, which further activates Akt and other kinases involved in cell growth and survival.

The importance of PIP3 in intracellular signal transduction makes it a target for many research and therapeutic interventions, especially in cancer research and treatment. PIK3, PTEN, and downstream factors such as Akt are crucial for the balance of PIP3. If they are disrupted, it can trigger a cascade reaction. In many cancers, mutations or overactivation of PI3K, and mutations or loss of the PTEN gene, lead to the accumulation of PIP3, resulting in persistent signal transduction that promotes tumor cell proliferation and survival. Therefore, various PI3K and Akt inhibitors have been used in clinical trials for cancer treatment. Therapies aimed at restoring or enhancing PTEN function are also being developed, all to balance PIP3 levels.

Is there any technical assistance where we can ask some questions before we order? Specifically, we are wondering about the solvent that we should use to re-solubilize the lipid after extraction.

Before using in the ELISA, the extracted lipid samples will be reconstituted in PBS-T 0.25%PS buffer which should be prepared with kit components according to the steps 1 and 2 under Reagent Preparation section on page 2 of the Data Sheet.
