General Notes
This product (Catalog No. DPABH-21996) is a rabbit-derived polyclonal antibody raised against human ST6GAL1, targeting the amino acid region 27–406 of the protein. It has been affinity purified by Protein A. Lactosylceramide alpha-2,6-N-sialyltransferase is a glycosyltransferase enzyme classified under EC 2.4.3.1 that catalyzes the transfer of N-acetylneuraminic acid (sialic acid) from the donor substrate CMP-N-acetylneuraminate to the non-reducing terminal β-D-galactosyl residue of lactosylceramide (LacCer; Galβ1-4Glcβ1-Cer) via an α-2,6 glycosidic linkage, yielding sialyl(α2,6)lac-tosylceramide and CMP. This sialylation step contributes to the biosynthesis of complex glycolipids, particularly minor ganglioside species with α2,6-linked sialic acid, which are less abundant compared to the predominant α2,3-linked structures like GM3. The enzyme's activity on LacCer has been documented in mammalian tissues such as rat liver and brain, where it participates in glycolipid modification alongside its primary role in protein glycosylation. Also known as β-galactoside α-2,6-sialyltransferase I (ST6Gal I), this enzyme is encoded by the ST6GAL1 gene in humans and exhibits broad substrate specificity for terminal β-galactosyl residues on both N-glycans of glycoproteins and select glycolipids. Encoded by a gene on chromosome 3q21.3, ST6Gal I is a type II transmembrane protein localized to the Golgi apparatus, where it plays a key role in terminal sialylation during the acute-phase response, immune cell function (e.g., generating sialylated IgG in B lymphocytes), and cell surface glycan diversity. In the context of glycolipids, its α2,6-sialyltransferase activity on LacCer is regulated by endogenous inhibitors, such as heat-stable acidic proteins identified in brain supernatants, which modulate both glycolipid and glycoprotein sialylation without affecting other glycosyltransferases.
DPABH-21996 is suitable for use in ELISA (1:4,000–1:8,000), IHC (1:50–1:100), Western blot (WB) (1:1,000–1:4,000) applications.
DPABH-21996 is supplied as a liquid formulation in 0.01M PBS, pH 7.4, 50% Glycerol, 0.05% Proclin 300. For long-term storage, keep at −20 °C. For short-term use, store at 2–8 °C. Avoid repeated freeze–thaw cycles to maintain antibody integrity and performance.
