This product (Catalog No. DPABH-28809) is a rabbit-derived polyclonal antibody raised against human ATP1A3, targeting the amino acid region 375–583 of the protein. It has been affinity purified by Protein A. ATP1A3 is a gene located on chromosome 19q13.2 that encodes the alpha-3 catalytic subunit of the Na⁺/K⁺-ATPase, a transmembrane ion pump essential for establishing and maintaining electrochemical gradients of sodium and potassium ions across cell membranes in neurons. This subunit, a 1,013-amino acid protein with seven transmembrane domains and a cytoplasmic catalytic site, forms heterooligomers with beta subunits to hydrolyze ATP and facilitate the active transport of three Na⁺ ions out of the cell in exchange for two K⁺ ions, thereby supporting membrane potential, osmotic balance, and neuronal excitability. Expressed predominantly in the central nervous system, including regions like the basal ganglia, hippocampus, and cerebellum, ATP1A3 plays a critical role in early brain development, particularly in cortical neurogenesis, neuronal migration, and the maturation of deep-layer excitatory neurons in the subplate during midgestation. Mutations in ATP1A3, mostly de novo heterozygous missense variants, disrupt pump function through loss-of-function mechanisms such as reduced ion affinity, impaired enzyme stability, or altered phosphorylation, leading to neuronal hyperexcitability and a spectrum of neurodevelopmental and neurodegenerative disorders.
DPABH-28809 is suitable for use in ELISA (1:4,000–1:8,000), IHC (1:50–1:100), Western blot (WB) (1:1,000–1:4,000) applications.
DPABH-28809 is supplied as a liquid formulation in 0.01M PBS, pH 7.4, 50% Glycerol, 0.05% Proclin 300. For long-term storage, keep at −20 °C. For short-term use, store at 2–8 °C. Avoid repeated freeze–thaw cycles to maintain antibody integrity and performance.
