BMP3 Full Name
bone morphogenetic protein 3
BMP3 Introduction
BMP3 (bone morphogenetic protein 3) is a secreted signaling molecule of the TGF-β superfamily that, unlike most BMPs, acts predominantly as a negative regulator of bone density. BMP3 signals through a heteromeric receptor complex composed of activin receptor type IIB (ACVR2B/ActR-IIB) and activin A receptor type I (ACVR1/ALK2), activating SMAD1/5/9 signaling but at a lower amplitude than osteogenic BMPs such as BMP2 and BMP7. The gene maps to chromosome 4q21.31 and is highly expressed in bone, lung, and intestinal tissues.

Negative Regulator of Bone Formation
BMP3 competes with osteogenic BMPs for ACVR2B binding and acts as a dominant-negative modulator of SMAD1/5/9 signaling in osteoblasts, restraining bone formation in response to mechanical loading and to systemic factors. Genetic ablation of BMP3 in mice produces high bone mass, while overexpression reduces trabecular bone volume. The BMP3 protein circulates at appreciable concentrations in plasma and acts as an endocrine 'osteokine' that links bone remodeling to systemic metabolism. BMP3 is also required for normal tooth development and for the patterning of the intestinal villus epithelium.
Bone Density Disorders and Regenerative Medicine
Common variants in BMP3 have been associated with low bone mineral density and with the risk of osteoporotic fractures in genome-wide association studies, supporting its role as a regulator of skeletal homeostasis. Recombinant BMP3 is being explored as an antagonist of excessive BMP signaling in fibrodysplasia ossificans progressiva, where it may limit heterotopic bone formation. In regenerative medicine, BMP3 supplementation has shown promise in promoting tendon-bone healing, while BMP3 antagonists such as Gremlin-1 are being developed to enhance bone formation in osteoporosis.
