Application Notes
The binding reactivity of this antibody towards Dxd-KLH and Dxd-BSA conjugates was determined using ELISA. This antibody is capable of binding antibody-drug conjugates ADC-8201 and ADC029 in a competitive ELISA, wherein the antibody was trastuzumab. The binding affinity of this antibody towards Dxd-BSA was determined using surface plasmon resonance (SPR) and it was reported to be Kd= 1.42nM
General Notes
The Hi-Puri™ Human Anti-DXD Monoclonal Antibody, HRP-conjugated (clone 14E3-1, product code CABT-L1916W) is a human monoclonal antibody specifically directed against DXD (Dxd), also known as an exatecan derivative or camptothecin analog. DXD is a potent cytotoxic small-molecule payload that acts as a DNA topoisomerase I (TOP1) inhibitor and is widely used in antibody-drug conjugate (ADC) development. By forming a stable ternary complex with TOP1 and DNA, DXD inhibits DNA replication, induces cell cycle arrest, and promotes cancer cell death. DXD-based ADCs, including HER2-targeting DXD conjugates, are extensively studied for targeted delivery of cytotoxic agents in oncology research. This antibody is suitable for research applications involving DXD detection, ADC characterization, and bioanalytical assay development.
This product is validated for use in ELISA applications and is conjugated with HRP for direct signal detection. The binding reactivity of this antibody toward DXD-KLH and DXD-BSA conjugates was determined using ELISA. The antibody is capable of recognizing DXD-containing antibody-drug conjugates, including ADC-8201 and ADC029, in competitive ELISA formats. The binding affinity toward DXD-BSA was characterized using surface plasmon resonance (SPR), with a reported affinity of Kd = 1.42 nM. This antibody specifically reacts with both DC-8201 and ADC029, enabling broad detection of DXD-containing ADC molecules.
CABT-L1916W is a human IgG monoclonal antibody purified by affinity purification and conjugated with HRP. It is supplied as a liquid formulation in PBS and available in multiple sizes, including 100 μg, 200 μg, 500 μg, and 1 mg. For long-term storage, the product should be stored at −20°C and repeated freeze–thaw cycles should be avoided. The product is shipped on dry ice. This product is intended for research use only and is not intended for diagnostic use.
