Intended Use

The Androstenedione ELISA Kit is an enzyme immunoassay for the quantitative measurement of Androstenedione in serum and EDTA plasma.

Contents of Kit

1. Microtiterwells: 12 × 8 (break apart strips), wells coated with anti-Androstenedione antibody (polyclonal). 96 (8 × 12) wells
2. Standards and Controls: ready to use
Standard A: 0 ng/mL, 1 mL
Standard B: 0.1 ng/mL, 1 mL
Standard C: 0.3 ng/mL, 1 mL
Standard D: 1.0 ng/mL, 1 mL
Standard E: 3.0 ng/mL, 1 mL
Standard F: 10 ng/mL, 1 mL
Control 1 (low): For control values and ranges please refer to vial label or QC-Report, 1 mL
Control 2 (high): For control values and ranges please refer to vial label or QC-Report, 1 mL
Contents: Contain non-mercury preservative. Standards are calibrated against Reference Material of the National Measurement Institute Australia (NMIA-M955).
Conversion: ng/mL x 3.492 = nmol/L
Note: Additional Standard A for sample dilution is available upon request.
3. Enzyme Conjugate: Ready to use, Androstenedione conjugated to horseradish peroxidase. Contains non-mercury preservative. 25 mL
4. Substrate Solution: Ready to use, tetramethylbenzidine (TMB). 25 mL
5. Stop Solution: Ready to use, contains 0.5 M H₂SO₄. Avoid contact with the stop solution. It may cause skin irritation and burns. 14 mL
6. Wash Solution: 40× concentrated. 30 mL

Storage

When store at 2°C to 8°C unopened reagents will retain reactivity until expiration date. Do not use reagent beyond this date.
Opened reagents must be stored at 2°C to 8°C. Microtiter wells must be stored at 2°C to 8°C. Once the foil bag has been opened, care should be taken to close it tightly again. Opened kits retain activity for 8 weeks if stored as described above.

Performance Characteristics

High-Dose-Hook Effect
Hook effect was not observed in this test up to a concentration of 186 ng/mL of Androstenedione.
Expected Normal Values
It is strongly recommended that each laboratory should determine its own normal and abnormal values.
In a study conducted with serum of apparently normal healthy donors, using the Androstenedione ELISA

Detection Range

The range of the assay is between 0.021 – 10 ng/mL.

Sensitivity

The analytical sensitivity of the Androstenedione ELISA Kit was calculated by subtracting 2 standard deviations from the mean of 20 replicate analyses of the Standard A and was found to be 0.021 ng/mL.
