The Melanocortin-4 receptor (MC4R) is a class A G-protein-coupled receptor (GPCR) encoded by an intronless gene on chromosome 18q21.3, consisting of 332 amino acids organized into seven transmembrane domains, and it serves as a key regulator of energy homeostasis, appetite control, and body weight balance primarily through its expression in hypothalamic neurons. Encoded by a 999-base-pair gene, MC4R exhibits high evolutionary conservation, with 93% amino acid identity between human and rat sequences, underscoring its fundamental physiological role across species.
