ELISA was used to demonstrate that Hfab-aNS3 binds to NS3 protein, with the relative affinity found to be in the range of 1 nmol/L. Through helicase assays it was established that Hfab-aNS3 blocks the helicase activity of NS3 protein. Intracellular expression of Hfab-aNS3 inhibits replication and expression of NS3 protein in a replicon cell line, as shown through immunocytochemistry.
