PTHrP intracrine actions divergently influence breast cancer growth through p27 and LIFR
Figure 1. Secreted PTHrP (1-34aa) levels measured by ELISA from conditioned media of cells described in (A). (B-E & G) n = 3 independent biological replicates. Graphs represent mean ± SEM. Parathyroid hormone-related protein (PTHrP) is a proteinaceous hormone that belongs to the parathyroid hormone (PTH) family. PTHrP is primarily secreted by mesenchymal stem cells, but it can also be produced by certain cancer cells, such as breast cancer and certain types of lung cancer. PTHrP plays a crucial role in regulating various physiological processes in the body. It acts as a versatile hormone, functioning as an endocrine, autocrine, paracrine, and intracrine signaling molecule. Its functions extend to different tissues and organs, including bone, teeth, vascular tissues, and mammary glands.
One of the key roles of PTHrP is in the regulation of bone development. It maintains the endochondral growth plate, which is responsible for longitudinal bone growth, at a constant width. PTHrP also influences epithelial-mesenchymal interactions during the formation of mammary glands, contributing to normal mammary gland development. Furthermore, PTHrP is involved in calcium homeostasis, a vital process in vertebrates. It helps regulate the levels of calcium in the body, ensuring its proper balance. PTHrP also has diverse functions in other systems. In the cardiovascular system, it acts as a vascular relaxant by affecting the smooth muscle of blood vessels. It also has variable effects on the heart, influencing its contractility over time. In the pancreas, PTHrP is involved in promoting the proliferation of β cells, which produce insulin, and inhibiting the apoptosis of these cells. In the central nervous system, PTHrP has a protective effect on neurons by safeguarding them from damage caused by excessive depolarization.
PTHrP is involved in the "cytokines excess" syndrome observed in cancer patients. It contributes to cancer cachexia by activating a gene program related to thermogenesis in brown adipose tissue. Unfortunately, blocking excessive cytokines or the PTH/PTHrP receptor does not alleviate cancer cachexia due to potential widespread side effects. However, the use of PTHrP as a reliable tumor marker in oncology is hindered by issues related to assay specificity, sensitivity, and analytical precision. Resolving these problems is crucial, especially considering that the function of PTHrP fragments formed by local proteolysis remains largely unknown.

Q: PTH-RP ( 1-34) ( human, rat, mouse) ELISA Kit (DEIA2034). There is also another ELISA kit for PTH 1-34 (DEIABL239). What is the difference between the two kits?
A: The detection targets are different.
DEIA2034: Detection of PTH-RP, which is a paracrine/autocrine/intracrine factor widely produced by many tissues of the body (breast, skin, blood vessels, placenta, fetal tissue, etc.). It plays many important roles in normal physiology, such as regulating chondrocyte differentiation (fetal bone development, growth plate), breast development, vascular tension, tooth eruption, skin hair growth, etc. It is not the main hormone for maintaining blood calcium homeostasis.
DEIABL239: Detection of PTH, which is synthesized and secreted by parathyroid chief cells. It is the main hormone for maintaining blood calcium homeostasis. Hypocalcemia stimulates its secretion, acting on bones and kidneys to increase blood calcium. Teriparatide is a synthetic drug (recombinant human PTH (1-34)). Its existence and function are entirely for therapeutic purposes (mainly osteoporosis).
