Intended Use
The Human IL1RL1 / DER4 ELISA Pair Set is for the quantitative determination of Human IL1RL1/DER5.
This ELISA Pair Set contains the basic components required for the development of sandwich ELISAs.
Contents of Kit
Capture Antibody: 1 mg/mL of mouse anti-Human IL1RL1 / DER4 monoclonal antibody (in PBS, pH 7.4). Dilute to a working concentration of 2 μg/mL in PBS before coating.
Detection Antibody: 0.2 mg/mL of mouse anti-Human IL1RL1 / DER4 monoclonal antibody conjugated to horseradish-peroxidase (HRP) (in PBS, 50 % HRP-Protector, pH 7.4, store at 4°C). Dilute to working concentration of 1.2 μg/mL in detection antibody dilution buffer before use.
Standard: Each vial contains 28 ng of recombinant Human IL1RL1 / DER4. Reconstitute with 1 mL detection antibody dilution buffer. After reconstitution, store at -20°C to -80°C in a manual defrost freezer. A seven-point standard curve using 2-fold serial dilutions in sample dilution buffer, and a high standard of 500 pg/mL is recommended.
Storage
Capture Antibody: Aliquot and store at -20°C to -80°C for up to 6 months from date of receipt. Avoid repeated freeze-thaw cycles.
Detection Antibody: Store at 4°C and protect it from prolonged exposure to light for up to 6 months from date of receipt. Do not freeze!
Standard: Store lyophilized standard at -20°C to -80°C for up to 6 months from date of receipt. Aliquot and store the reconstituted standard at -80°C for up to 1 month. Avoid repeated freezethaw cycles.
Sensitivity
The minimum detectable dose of Human IL1RL1 / DER4 was determined to be approximately 7.8 pg/ml.
This is defined as at least three times standard deviations above the mean optical density of 10 replicates of the zero standard.
General Description
Interleukin-1 receptor-like 1, also known as protein ST2, IL1RL1 and DER4, is a single-pass type I membrane and a secreted protein which belongs to the interleukin-1 receptor family. IL1RL1 / DER4 has a soluble secreted form (sST2) and a transmembrane form (ST2L). IL1RL1 / DER4 contains three Ig-like C2-type (immunoglobulin-like) domains and one TIR domain. IL1RL1 / DER4 is highly expressed in kidney, lung, placenta, stomach, skeletal muscle, colon and small intestine. Isoform A of IL1RL1 is prevalently expressed in the lung, testis, placenta, stomach and colon. Isoform B of IL1RL1 is more abundant in the brain, kidney and the liver. Isoform C of IL1RL1 is not detected in brain, heart, liver, kidney and skeletal muscle. The coding gene forms a cytokine receptor gene cluster together with IL1R1(interleukin 1 receptor, type I), IL1R2 (interleukin 1 receptor, type II) and IL1RL2 (interleukin 1 receptor-like 2) genes in a region mapped to chromosome 2q12. As an important mediator involved in many immune and inflammatory responses, this cytokine has been implicated as a regulator of both the development and effector phases of type 2 helper T cell responses, and as a negative feedback modulator of macrophage pro-inflammatory function. IL33 is a specific ligand of the protein IL1RL1 and induces production of Th2 cytokines.
