General Notes
This product (Catalog No. DPABH-16328) is a rabbit-derived IgG polyclonal antibody raised against human TIMELESS. This antibody exhibits cross-reactivity with TIMELESS from mouse. It has been affinity purified by protein A. The Timeless gene, often abbreviated as tim, encodes the TIMELESS (TIM) protein, a key regulator of circadian rhythms in animals, most notably in the fruit fly Drosophila melanogaster, where it forms a heterodimer with the PERIOD (PER) protein to inhibit CLOCK-CYCLE-driven transcription in the core feedback loop of the biological clock. Discovered in 1994 through genetic screens for mutants disrupting locomotor activity rhythms, the gene's product accumulates in the cytoplasm at night, facilitating PER's nuclear entry for repression, and undergoes light-induced degradation via CRYPTOCHROME (CRY) to enable daily clock resetting. In mammals, the orthologous TIMELESS gene retains roles in DNA replication fork stability and genomic integrity as part of the TIM-TIPIN complex, though its direct involvement in the suprachiasmatic nucleus clock is less central, with CRY proteins largely supplanting TIM's repressive function.
DPABH-16328 is suitable for use in Western blot (WB) (1:500–1:2,000), IHC (1:50–1:200) applications.
DPABH-16328 is supplied as a liquid formulation in PBS with 0.02% sodium azide, 50% glycerol, pH7.3. For long-term storage, keep at −20 °C. For short-term use, store at 2–8 °C. Avoid repeated freeze–thaw cycles to maintain antibody integrity and performance.
