This product (Catalog No. DPABH-18617) is a rabbit-derived polyclonal antibody raised against human PLN, targeting the amino acid region 1–52 of the protein. This antibody exhibits cross-reactivity with Human PLN from mouse and rat. It has been affinity purified by Protein A. Cardiac phospholamban (PLN) is a small regulatory phosphoprotein located on the membrane of the sarcoplasmic reticulum in cardiomyocytes, where it plays a key role in controlling intracellular calcium cycling and myocardial contractility. Encoded by the PLN gene, phospholamban inhibits the activity of the sarcoplasmic/endoplasmic reticulum Ca²⁺-ATPase (SERCA2a) under dephosphorylated conditions, thereby reducing calcium reuptake into the sarcoplasmic reticulum and decreasing cardiac muscle relaxation rate. Upon β-adrenergic stimulation, phospholamban is phosphorylated by protein kinase A or other kinases, relieving its inhibitory effect on SERCA2a and enhancing calcium sequestration, which promotes efficient cardiac relaxation and increases contractile performance. Mutations in PLN are associated with cardiomyopathies, including dilated and arrhythmogenic cardiomyopathy, highlighting its importance in cardiac excitation–contraction coupling and heart function regulation.
DPABH-18617 is suitable for use in ELISA (1:4,000–1:8,000), IHC (1:50–1:100), Western blot (WB) (1:1,000–1:4,000) applications.
DPABH-18617 is supplied as a liquid formulation in 0.01M PBS, pH 7.4, 50% Glycerol, 0.05% Proclin 300. For long-term storage, keep at −20 °C. For short-term use, store at 2–8 °C. Avoid repeated freeze–thaw cycles to maintain antibody integrity and performance.
