General Notes
This product (Catalog No. DPABH-25516) is a rabbit-derived polyclonal antibody raised against human GPR35. This antibody exhibits cross-reactivity with GPR35 from mouse. It has been affinity purified by Protein A. PR35 is a class A G protein-coupled receptor (GPCR) encoded by the GPR35 gene located on human chromosome 2q37.3, featuring seven transmembrane domains typical of GPCRs and serving as a key mediator in immune and inflammatory processes through its coupling to Gα12/13 and Gαi/o proteins. Discovered in 1998, GPR35 was initially identified as an orphan receptor, but subsequent research has established multiple endogenous ligands, including the chemokine CXCL17 (which induces chemotaxis in monocytes), kynurenic acid (an anti-inflammatory metabolite with micromolar potency), 5-hydroxyindoleacetic acid (promoting neutrophil recruitment), and lysophosphatidic acid (linked to pro-inflammatory effects). These ligands activate diverse downstream pathways in GPR35, such as MAPK/ERK, NF-κB, PI3K/AKT, and Rho-mediated cytoskeletal reorganization, often in a biased manner that can influence β-arrestin recruitment and receptor internalization. Synthetic agonists like zaprinast and pamoic acid, along with approved drugs such as lodoxamide and cromolyn disodium (mast cell stabilizers), further highlight its pharmacological profile, though ligand potency varies by species and isoform (GPR35a and GPR35b). GPR35 exhibits broad expression across tissues, with highest levels in the gastrointestinal tract (small intestine, colon, and stomach), immune cells (including monocytes, macrophages, neutrophils, T cells, and dendritic cells), and lower abundance in the liver, spleen, kidney, and lung. Its expression is upregulated during inflammation, microbial invasion, or IgE stimulation in contexts like allergic responses and infections, reflecting its role in mucosal immunity and barrier integrity.
DPABH-25516 is suitable for use in Western blot (WB) (1:500–1:2,000), IF (immunofluorescence) (1:200–1:1,000), ELISA (1:20000) applications.
DPABH-25516 is supplied as a liquid formulation in PBS containing 50% glycerol, 0.5% BSA and 0.02% sodium azide. For long-term storage, keep at −20 °C. For short-term use, store at 2–8 °C. Avoid repeated freeze–thaw cycles to maintain antibody integrity and performance.
