Drosha is a member of the ribonuclease (RNase) III family. Like other RNase III proteins, Drosha is a double-stranded RNA (dsRNA)-specific endonuclease that introduces staggered cuts on each strand of the RNA helix. The human genome encodes two RNase III proteins: Drosha and Dicer. Dicer homologs are found in a variety of eukaryotic organisms. Drosha homologs are present only in animals but not in yeast or plants. Drosha plays a key role in the generation of precursor microRNAs from primary microRNA (pri-miRNA) transcripts in animal cells, but how it recognizes its RNA substrates is not fully understood. It is widely thought that Drosha is directly involved in executing the initial step of microRNA processing in the nucleus, that is cleavage of pri-miRNA to release pre-miRNApre-rRNA.
