This product (Catalog No. DPABH-22638) is a rabbit-derived polyclonal antibody raised against human DLAT, targeting the amino acid region 1–322 of the protein. This antibody exhibits cross-reactivity with Human DLAT from mouse and rat. It has been affinity purified by Protein A. This gene encodes component E2 of the multi-enzyme pyruvate dehydrogenase complex (PDC). PDC resides in the inner mitochondrial membrane and catalyzes the conversion of pyruvate to acetyl coenzyme A. The protein product of this gene, dihydrolipoamide acetyltransferase, accepts acetyl groups formed by the oxidative decarboxylation of pyruvate and transfers them to coenzyme A. Dihydrolipoamide acetyltransferase is the antigen for antimitochondrial antibodies. These autoantibodies are present in nearly 95% of patients with the autoimmune liver disease primary biliary cirrhosis (PBC). In PBC, activated T lymphocytes attack and destroy epithelial cells in the bile duct where this protein is abnormally distributed and overexpressed. PBC enventually leads to cirrhosis and liver failure. Mutations in this gene are also a cause of pyruvate dehydrogenase E2 deficiency which causes primary lactic acidosis in infancy and early childhood.[provided by RefSeq, Oct 2009].
DPABH-22638 is suitable for use in Western blot (WB) (1:2,000–1:10,000), IP (0.5-4.0 μg for 1.0-3.0 mg of total protein lysate), IHC (1:500–1:2,000), IF/ICC (1:200–1:800) applications.
DPABH-22638 is supplied as a liquid formulation in PBS with 0.02% sodium azide and 50% glycerol, pH 7.3. For long-term storage, keep at −20 °C. For short-term use, store at 2–8 °C. Avoid repeated freeze–thaw cycles to maintain antibody integrity and performance.
