Tnfsf8 Introduction
TNFSF8 (Tumor Necrosis Factor Superfamily Member 8), also widely known as CD30 ligand (CD30L) or CD153, is a cytokine-encoding gene located on human chromosome 9q32-q33.1. It is a protein-coding gene with a pLI score of 0.96, indicating strong intolerance to loss-of-function variation, and a LOEUF score of 0.51. The gene spans several exons and produces multiple transcript variants encoding different isoforms. TNFSF8 is highly conserved across vertebrates, with clear orthologs identified in mouse (Tnfsf8), rat (Tnfsf8), and even Xenopus tropicalis, highlighting its fundamental biological importance.

Protein Structure and Domain Architecture
TNFSF8 encodes a type II transmembrane protein of approximately 26-30 kDa that belongs to the tumor necrosis factor (TNF) ligand family. The protein exhibits a characteristic three-domain architecture: a short N-terminal cytoplasmic tail involved in intracellular trafficking, a single transmembrane helix that anchors the protein within the cell membrane, and an extracellular C-terminal domain responsible for receptor binding. The extracellular region contains a conserved TNF homology domain that adopts a characteristic "jelly-roll" beta-sandwich fold, enabling specific interaction with its cognate receptor, CD30/TNFRSF8. TNFSF8 can exist in both membrane-bound and soluble forms, with the latter generated through proteolytic cleavage, and both variants retain the ability to bind CD30.
Immune Regulatory Functions and Signaling Mechanisms
TNFSF8 functions as a critical immune modulator through its exclusive interaction with CD30, a receptor expressed on activated T and B lymphocytes. Upon engagement, TNFSF8-CD30 signaling regulates diverse immune processes including T-cell activation, proliferation, and differentiation, particularly promoting T helper 2 (Th2) cell responses essential for humoral immunity. The ligand-receptor interaction also enhances B-cell activation, antibody production, and immunoglobulin class switching, contributing to adaptive immune responses. Notably, TNFSF8 exhibits pleiotropic and context-dependent effects on target cells: in some lymphoma cell lines it promotes proliferation, while in others it induces cell death and reduces growth. The pathway also modulates CD8-positive T-cell differentiation and plays roles in defense against Gram-positive bacteria.
