PRKCZ Full Name
protein kinase C, zeta
PRKCZ Introduction
Protein kinase C zeta (PRKCZ, or PKCζ) is a highly specialized enzyme that belongs to the "atypical" protein kinase C (aPKC) subfamily. Unlike conventional and novel PKCs, the structure of PRKCZ lacks the regulatory domains required for binding calcium ions and diacylglycerol (DAG). Consequently, its enzymatic activation is entirely independent of both calcium and DAG. Instead, PRKCZ is primarily activated downstream of phosphoinositide 3-kinase (PI3K) through the generation of lipid second messengers like phosphatidylinositol 3,4,5-trisphosphate (PIP3), as well as through highly specific protein-protein interactions.

PRKCZ functions as a critical molecular signaling hub across multiple fundamental cellular pathways. One of its most universally recognized roles is in the establishment and maintenance of cell polarity, which is achieved through its physical association with the PAR (partitioning defective) protein complex. This function is absolutely vital for proper tissue architecture, especially in epithelial cells. Beyond cellular organization, PRKCZ is an indispensable downstream effector in the insulin signaling pathway. Upon insulin receptor activation, PRKCZ mediates the translocation of GLUT4 glucose transporters to the plasma membrane, facilitating cellular glucose uptake in key metabolic tissues like skeletal muscle and adipose tissue. Additionally, it plays a prominent role in innate immunity and cellular survival by participating in the activation of the NF-κB transcription factor cascade. Because of its central position in both metabolic and architectural signaling, the dysregulation of PRKCZ is deeply linked to two major global health burdens: metabolic disorders and cancer. In the context of metabolic syndrome, a decline in PRKCZ activity or its impaired activation by insulin is a major, well-documented molecular defect driving insulin resistance and the subsequent development of type 2 diabetes.
In oncology, the role of PRKCZ is highly complex and heavily tissue-dependent. While it can promote survival in certain contexts, accumulating evidence strongly suggests that PRKCZ predominantly acts as a critical tumor suppressor in several severe malignancies, including colorectal and prostate cancers. The genetic loss or functional downregulation of PRKCZ in these epithelial cancers disrupts cell polarity, dismantles normal tissue architecture, and actively drives epithelial-to-mesenchymal transition (EMT), thereby heavily promoting tumor invasion and distant metastasis.
