GRAP2
GRAP2 Full Name
GRB2-related adaptor protein 2
GRAP2 Introduction
GRAP2 (GRB2-related adaptor protein 2), also called GADS, is a hematopoietic-specific adaptor protein that links T-cell receptor engagement to downstream signaling cascades. Its SH3–SH2–SH3 architecture closely mirrors that of GRB2, but a unique 50-residue insertion between the two SH3 domains provides a docking site for the adaptor SLP-76 and biases the protein toward lymphoid signaling contexts. The gene maps to chromosome 22q13.1, and expression is largely confined to T cells, NK cells, and certain myeloid lineages.

Coupling the T-Cell Receptor to SLP-76 and LAT
Upon TCR stimulation, the SH2 domain of GRAP2 binds phosphorylated LAT, while its central SH3 domain recruits the adaptor SLP-76, which in turn brings together Vav1, Nck, and PLCγ1 to form a productive signaling cluster at the immunological synapse. The extended linker region of GRAP2 inserts between LAT and SLP-76 and is essential for proper signaling output: without it, the LAT–SLP-76 complex is destabilized and downstream calcium flux, NFAT activation, and ERK phosphorylation are blunted. GRAP2 also contributes to CXCR4-driven chemokine responses in developing thymocytes and to NK cell education through its interactions with members of the SLAM family.
T-Cell Development and Immune Dysregulation
GRAP2-deficient mice exhibit a partial block at the double-positive to single-positive transition in the thymus, and residual mature T cells show impaired proliferation and cytokine production. In humans, rare loss-of-function variants have been described in patients with combined immunodeficiency featuring recurrent viral and bacterial infections. Genetic and pharmacological studies have highlighted GRAP2 as a candidate target in autoimmune disease: disruption of the GRAP2–SLP-76 interaction dampens pathogenic T-cell responses in models of graft-versus-host disease, motivating the development of small-molecule GRAP2–SLP-76 disruptors as next-generation immunomodulators.
