BMP4 Introduction
BMP4 (Bone Morphogenetic Protein 4) encodes a secreted ligand belonging to the transforming growth factor-beta (TGF-β) superfamily, located on chromosome 14q22.2. The gene produces a 408-amino acid preproprotein that undergoes proteolytic processing to generate the mature, disulfide-linked homodimer. Structurally, the protein contains a TGF-beta propeptide domain and a conserved TGFB domain that mediates receptor binding and dimerization. BMP4 is also known by the synonyms BMP-2B and bone morphogenetic protein 2B, reflecting its close evolutionary relationship with other BMP family members.

Essential Functions in Embryonic Development
BMP4 serves as a master regulator of vertebrate embryogenesis, controlling critical processes including mesoderm formation, neurogenesis, and organ morphogenesis. During early development, BMP4 directs the specification of ventral-caudal structures and establishes body axes, functioning as an organizer for posterior tissue formation. The protein exhibits broad expression in developing organs including heart, lung, liver, kidney, limbs, and nervous system. Notably, asymmetric expression of Bmp4 in the gut mesogastrium precedes gastrointestinal organ asymmetry, highlighting its role in left-right patterning. In later development, BMP4 regulates tooth morphogenesis, osteoblast differentiation, and mammary gland ductal outgrowth.
Signal Transduction Mechanisms
BMP4 initiates cellular responses through both canonical and non-canonical signaling pathways. In the canonical cascade, BMP4 binds cooperatively to type I (BMPR1A) and type II (BMPR2) serine/threonine kinase receptors, forming a ternary complex at the cell surface. BMPR2 then phosphorylates and activates BMPR1A, which propagates the signal by phosphorylating SMAD1/5/8 transcription factors. These phosphorylated SMADs translocate to the nucleus and regulate target gene expression. Additionally, BMP4 activates non-canonical pathways including ERK/MAP kinase, PI3K/Akt, and SRC cascades, with SRC-mediated VEGFR2 activation promoting angiogenic responses.
