APOE Introduction
The liver and brain and macrophages produce Apolipoprotein E (APOE) which functions as a lipid-binding protein to manage lipoprotein metabolism and cholesterol transport. The protein links to chylomicrons and VLDL and HDL particles which allows tissue lipid distribution. The protein APOE functions through its ability to bind with LDL receptor family members which enables the removal of triglyceride-rich lipoprotein remnants from blood circulation. The three human APOE isoforms APOE2 and APOE3 and APOE4 show different biochemical properties because of their single amino acid variation yet they have distinct clinical effects.

APOE functions as an essential protein which regulates lipid equilibrium and enables neuronal membrane restoration and controls inflammatory reactions. The central nervous system depends on APOE as its primary apolipoprotein which astrocytes produce to deliver cholesterol to neurons and maintain synaptic structure and repair damaged tissue. The immune system responds to APOE through its ability to control both pro-inflammatory and anti-inflammatory signaling pathways. The different APOE isoforms produce distinct effects on lipid metabolism and neuronal function and systemic inflammation because they bind receptors differently and stabilize lipids and structures at different levels.
The medical field recognizes APOE as the gene which strongly links to Alzheimer's disease because APOE4 carriers face higher disease risk and develop symptoms earlier but APOE2 carriers seem to have protective benefits. The metabolic and cardiovascular disorders type III hyperlipoproteinemia and atherosclerosis and coronary artery disease develop because of APOE functional disturbances. The APOE4 allele results in inadequate removal of amyloid-β and causes tau protein damage and brain inflammation which leads to neurodegenerative diseases. The APOE gene variants link to three additional conditions which include cerebrovascular disease and traumatic brain injury results and immune system disorders that demonstrate its wide-ranging effects on metabolic and neurological and inflammatory systems.
