After you receive the kit, all the components should be stored in the refrigerator (4-8°C) also up to 18 months. Long term storage, improper storage conditions and large temperature fluctuation cycles may cause precipitates in the TMB solution. These precipitates should not affect the assay noticeably. Nevertheless, if you observe such precipitates, we recommend to avoid them by allowing them to sink to the bottom.

General Description
Semaglutide is a therapeutically effective analog of glucagon-like peptide-1 (GLP-1). GLP-1 is an incretin hormone with important effects on glycemic control and body weight regulation, particularly relevant in people with type 2 diabetes (T2D). This ELISA was developed with serum from rabbits immunized with Semaglutide coupled to a carrier protein. Background
Type 2 diabetes mellitus (T2DM) refers to a metabolic disease in which the body's insulin deficiency or insulin utilization is reduced, resulting in a sustained increase in blood sugar in patients. Patients are often accompanied by abnormal lipid metabolism, and most patients are accompanied by obesity. Long-term exposure of the body to high blood sugar and high fat environments can lead to lesions in blood vessels and nerves, which can harm the heart, kidneys and other organs. Metformin is one of the most commonly used drugs to treat T2DM, but long-term use of metformin alone will cause a series of complications. This shows that simply increasing the dose of hypoglycemic drugs can no longer effectively improve the therapeutic effect of T2DM patients. Therefore, in the treatment of T2DM patients, the issue of obesity also needs to be paid attention. Studies have found that glucagon-like peptide-1 receptor agonist (GLP-1RA) is an effective drug to achieve good blood sugar control in patients with type 2 diabetes (T2DM). GLP-1RA not only lowers blood sugar but also has weight loss effects.

Semaglutide is a new generation of GLP-1 (glucagon-like peptide-1) analogue. It is a long-acting dosage form developed based on the basic structure of liraglutide. It is more effective in treating type 2 diabetes. As a glucagon-like peptide 1 receptor agonist, semaglutide increases the production of insulin, a hormone that lowers blood sugar, by mimicking the effects of glucagon-like peptide-1 (GLP-1). It also appears to promote the growth of pancreatic cells, which are responsible for the production and release of insulin. Additionally, it inhibits the production of glucagon, a hormone that promotes glycogenolysis (the release of stored carbohydrates from the liver) and gluconeogenesis (the synthesis of new glucose). It reduces food intake by reducing appetite and slowing digestion in the stomach, helping to reduce body fat.
Because semaglutide not only has the function of lowering blood sugar and lipids, it also shows efficacy and effects on immune regulation, antioxidant, and promotion of growth and development. Therefore, researchers are still actively exploring its therapeutic effects on other diseases. Semaglutide is a glucagon-like peptide-1 receptor agonist that was recently approved by the US Food and Drug Administration for chronic weight management. This paper reviews data on the mechanism of action, weight-loss and cardiometabolic efficacy, and safety of semaglutide 2.4 mg/week for obesity. Semaglutide has demonstrated the largest weight loss of any obesity medication to date with reductions of approximately 15% of initial weight at 68 weeks, accompanied by improvements in cardiovascular risks factors and physical functioning. The approval of this medication provides patients with greater options for weight management.
Wegovy (semaglutide): a new weight loss drug for chronic weight management
Obesity is a growing epidemic within the USA. Because weight gain is associated with an increased risk of developing life-threatening comorbidities, such as hypertension or type 2 diabetes, there is great interest in developing non-invasive pharmacotherapeutics to help combat obesity. Glucagon-like peptide-1 (GLP-1) receptor agonists are a class of antidiabetic medications that have shown promise in encouraging glycemic control and promoting weight loss in patients with or without type 2 diabetes. This literature review summarizes and discusses the weight loss results from the SUSTAIN (Semaglutide Unabated Sustainability in Treatment of Type 2 Diabetes), PIONEER (Peptide Innovation for Early Diabetes Treatment), and STEP (Semaglutide Treatment Effect in People with Obesity) clinical trial programs. The SUSTAIN and PIONEER clinical trials studied the use of 1.0 mg, once-weekly, subcutaneous and oral semaglutide (a new GLP-1 homolog), respectively, on participants with type 2 diabetes. The STEP trial examined the effects of 2.4 mg, once-weekly, subcutaneous semaglutide on patients with obesity. Trial data and other pertinent articles were obtained via database search through the US National Library of Medicine Clinical Trials and the National Center for Biotechnology Information. All three clinical trials demonstrated that semaglutide (injected or oral) has superior efficacy compared with placebo and other antidiabetic medications in weight reduction, which led to Food and Drug Administration approval of Wegovy (semaglutide) for weight loss.
