The antigen is prepared by inoculating the standard strain of Salmonella pullorum C79-1 (CVCC 79201) and the variant strain of Salmonella gallinarum C79-7 (CVCC 79207) into a suitable culture medium, harvesting the culture, mixing and then processing with a phosphate buffer solution containing formaldehyde, followed by ethanol treatment, and the addition of crystalline violet ethanol solution and glycerol.

Introduction
Salmonella pullorum and Salmonella gallinarum are closely related, host-adapted serovars of Salmonella enterica that primarily infect poultry. S. pullorum is the causative agent of Pullorum disease, a systemic infection in young chicks characterized by high mortality, while S. gallinarum causes fowl typhoid, an acute or chronic septicemic disease in adult birds. Both pathogens are non-motile and belong to the Salmonella enterica serogroup D, and they pose significant threats to poultry health and productivity, particularly in regions lacking effective control programs.
