General Description
H3F3A encodes histone H3.3, a replication-independent H3 variant deposited throughout the cell cycle at active genes, regulatory elements, and telomeres by dedicated chaperone pathways. It is central to chromatin dynamics, transcription, and epigenetic memory. Mutations in H3F3A (e.g., glioma-associated H3.3 K27M/G34) are established oncogenic drivers and clinically actionable diagnostic/prognostic markers, and H3.3 neoepitopes are under investigation as immunotherapy targets.
