GeneralInformation
Immunogen
Ebv-transformed b cells from a patient with mhc class-ii deficiency re-expressing class-ii antigens
Fusion Species
Mouse X Mouse Hybridoma
Immunological Donor
Mouse spleen
Cell Line Description
Described as secreting a mouse monoclonal antibody (igg1) CD39; the cell line was established in 1988 by fusion of PX63AG8.653 cells with spleen cells of a mouse immunized with EBV-transformed B cells from a patient with MHC class-II deficiency re-expressing class-II antigens .
Viruses: ELISA: reverse transcriptase negative; PCR: SMRV
Morphology
round, single cells growing in suspension and some slightly adherent
Growth Properties
suspension and some slightly adherent
CultureMethod
Subculturing
Split ratio: 1: 2 to 1: 4, every 2 days; Maintain at 0.2-0.4 x 10^6 cells/ml; Doubling time: 25 hours; Harvest: about 0.5 x 10^6 cells/ml.
Culture cells for the first 5-7 days in a 12-well-plate with HFCS, then cells may be transfered to flasks; after 14 days, cells become slightly adherent (about two-thirds of them), but can be detached by slapping the flask; the HFCS concentration can be decreased after the initial 2 weeks to about 50% (1: 100) or 25% (1: 200).
Storage
Liquid nitrogen vapor phase.
Freezing medium: to complete growth medium(20%FBS), add 10%(v/v) DMSO
