Leptin is a key adipokine linking adipose tissue mass to the regulation of appetite and energy expenditure, while coordinating broader physiological processes including neuroendocrine signaling, inflammation, and immunity. Beyond its classical role in body-weight homeostasis, leptin functions as an integrative regulator and a clinically relevant biomarker. Circulating leptin levels reflect not only fat mass but also sex, nutritional status, circadian rhythm, and disease context, supporting its use in studies of obesity, insulin resistance, cachexia, and chronic inflammation.
Leptin is a 16 kDa, non-glycosylated protein belonging to the long-chain helical cytokine family, characterized by a conserved four-helix bundle structure.
Leptin signals through the leptin receptor (Ob-R), with the long isoform Ob-Rb mediating full intracellular activity. Ligand binding induces receptor dimerization and activates JAK2, leading to downstream signaling via key pathways:

Signal intensity is tightly controlled by negative regulators such as SOCS3 and PTP1B, whose dysregulation contributes to leptin resistance.
Secreted primarily by white adipocytes in proportion to fat mass, leptin acts on hypothalamic nuclei, particularly the arcuate nucleus, to suppress appetite and increase energy expenditure. It is a critical determinant of body weight regulation and metabolic balance. However, the physiological role of leptin extends substantially beyond its canonical function in appetite control.
Beyond its mechanistic significance, leptin has increasingly been recognized as a clinically informative biomarker with relevance across a spectrum of metabolic and endocrine disorders. Circulating leptin concentrations provide an integrated readout of energy stores and metabolic status, enabling stratification of patients not only by adiposity but also by underlying pathophysiological states.

Unlike many classical biomarkers with relatively stable baseline levels, leptin exhibits substantial inter- and intra-individual variability driven by physiological, environmental, and pathological factors. Circulating concentrations span several orders of magnitude—from sub-ng/mL levels in lean or cachectic individuals to tens or even hundreds of ng/mL in obese populations. Consequently, robust leptin detection demands reagents and assay architectures that can accommodate wide concentration ranges, resist matrix interference, and maintain epitope fidelity under assay conditions.
Immunoassay-based methodologies remain the dominant analytical framework for leptin quantification, owing to their sensitivity, scalability, and compatibility with complex biological matrices. Among these, sandwich immunoassays represent the prevailing format. Sandwich -Enzyme-linked immunosorbent assays (s-ELISA) remain the most widely adopted platform, offering robustness and ease of implementation. Within the framework of sandwich immunoassays, the quality and compatibility of antibody pairs represent the primary determinants of analytical performance.
Creative Diagnostics provides a suite of anti-leptin matched antibody pairs specifically optimized for immunoassay applications. Each pair is functionally validated in sandwich assay formats, enabling good performance. We also offers a comprehensive portfolio of leptin-related antibody reagents, suitable for ELISA, WB, IHC, and other immunological applications. These reagents are characterized for consistency and performance, providing researchers with reliable tools for detection and mechanistic studies of leptin signaling.
