Adeno-associated virus serotype 6 (AAV6) has emerged as a highly versatile and promising viral vector for human gene therapy. Distinguished by its unique ability to efficiently transduce a wide range of target tissues—including airway epithelium, skeletal muscle, cardiac muscle, and hematopoietic stem cells—AAV6 offers distinct advantages over other serotypes. Notably, AAV6 is one of the few AAV serotypes capable of efficiently transducing both murine and human hematopoietic stem cells, making it particularly valuable for ex vivo gene therapy applications. Additionally, AAV6 exhibits robust transduction of airway epithelial cells, positioning it as a leading candidate for gene therapy of cystic fibrosis and other respiratory disorders.Despite its promise, the successful development and clinical translation of AAV6-based gene therapies depend critically on accurate monitoring of host immune responses against the vector. Preexisting neutralizing antibodies (NAbs) and treatment-emergent anti-AAV6 antibodies can significantly reduce transduction efficiency, limit therapeutic efficacy, and pose patient safety risks. Reliable serological detection of anti-AAV6 antibodies is therefore essential for patient screening, dose stratification, and post-treatment monitoring. This page provides a comprehensive overview of AAV6 biology, its applications in gene therapy, and validated detection solutions including human and mouse anti-AAV6 antibody ELISA kits.

AAV6 utilizes multiple cellular receptors for attachment and entry, contributing to its broad tropism. The primary attachment receptor for AAV6 is heparan sulfate proteoglycan (HSPG), a feature shared with AAV2 but not with all serotypes. The interaction between AAV6 capsid and HSPG is mediated by specific basic amino acid residues on the capsid surface, including arginine and lysine residues at positions 531, 532, and 533. Following initial attachment, AAV6 engages co-receptors including epidermal growth factor receptor (EGFR) and platelet-derived growth factor receptor (PDGFR) to facilitate internalization via clathrin-mediated endocytosis.
| Target Tissue | Transduction Efficiency | Key Applications |
| Airway epithelium | High | Cystic fibrosis, COPD |
| Skeletal muscle | High | Muscular dystrophy |
| Cardiac muscle | Moderate | Heart failure, cardiomyopathy |
| Hematopoietic stem cells | High (unique among AAVs) | Ex vivo gene therapy |
| Liver | Moderate | Hemophilia, metabolic disorders |
| Lung | High | Pulmonary diseases |
| Cystic Fibrosis | Ex Vivo HSC Gene Therapy | Muscle-Directed Gene Therapy |
| AAV6 has been extensively studied for cystic fibrosis gene therapy due to its natural tropism for airway epithelial cells. Unlike AAV5, which also transduces the lung, AAV6 achieves more efficient transduction of the apical surface of airway epithelium, the primary site of pathology in cystic fibrosis. Clinical trials have evaluated AAV6 vectors encoding the cystic fibrosis transmembrane conductance regulator (CFTR) gene, demonstrating safety and partial efficacy. | AAV6 is one of the few AAV serotypes capable of efficiently transducing human CD34+ hematopoietic stem cells. This property has been exploited for ex vivo gene therapy applications targeting hematologic disorders including sickle cell disease, beta-thalassemia, and primary immunodeficiencies. Unlike lentiviral vectors, AAV6 offers a non-integrating platform with a favorable safety profile, though transduction efficiency remains an area of active optimization. | AAV6 exhibits high transduction efficiency in skeletal and cardiac muscle, making it attractive for muscular dystrophies. Studies in animal models have demonstrated sustained transgene expression following intramuscular or systemic administration of AAV6 vectors. The lower seroprevalence of neutralizing antibodies against AAV6 compared to AAV2 in human populations further supports its clinical utility. |
The Human Anti-AAV6 Antibody ELISA Kit is designed for the quantitative or qualitative determination of total anti-AAV6 antibodies (IgG) in human serum, plasma, or other biological fluids. This kit is optimized for high-throughput screening applications in clinical research and regulatory-compliant environments.
The Mouse Anti-AAV6 Antibody ELISA Kit is specifically designed for preclinical gene therapy studies using murine models. This kit enables researchers to assess the immunogenicity of AAV6-based vectors in mice, a critical parameter for vector optimization and dose selection.
AAV6 has emerged as a highly versatile and promising vector for human gene therapy, with unique capabilities for transducing airway epithelium, skeletal and cardiac muscle, and hematopoietic stem cells. However, the presence of preexisting neutralizing antibodies and the potential for treatment-emergent immune responses pose significant challenges to the successful clinical application of AAV6-based therapeutics. The Human Anti-AAV6 Antibody ELISA Kit and Mouse Anti-AAV6 Antibody ELISA Kit provide validated, ready-to-use solutions for serological screening and immunogenicity monitoring, offering high sensitivity, specificity (no cross-reactivity with other AAV serotypes), and reproducibility. These tools enable accurate patient selection, dose optimization, and longitudinal monitoring of immune responses, supporting the development of safer and more effective AAV6-based gene therapies. As the field continues to advance toward clinical applications for cystic fibrosis, muscular dystrophy, and hematologic disorders, reliable anti-AAV6 serology will remain an essential component of successful gene therapy programs.
