Intended Use
The Human Lipoprotein A ELISA (Enzyme-Linked Immunosorbent Assay) Kit is designed for detection of Lp(a) in human plasma, serum, milk, urine, and CSF samples.
Contents of Kit
1. Human Lp(a) Microplate: A 96-well polystyrene microplate (12 strips of 8 wells) coated with a polyclonal antibody against human Lp(a). 2-8°C
2. Sealing Tapes: Each kit contains 3 precut, pressure sensitive sealing tapes that can be cut to fit the format of the individual assay. 2-8°C
3. Human Lp(a) Standard: Human Lp(a) in a buffered protein base (60 ng, lyophilized). 2-8°C
4. Biotinylated Human Lp(a) Antibody (50×): A 50-fold concentrated biotinylated polyclonal antibody against human Lp(a)[120 μl]. -20°C
5. EIA Diluent Concentrate (10×): A 10-fold concentrated buffered protein base (30 ml). 2-8°C
6. Wash Buffer Concentrate (20×): A 20-fold concentrated buffered surfactant (30 ml, 2 bottles). 2-8°C
7. SP Conjugate (100×): A 100-fold concentrate (80 μl). -20°C
8. Chromogen Substrate (1×): A stabilized peroxidase chromogen substrate tetramethylbenzidine (7 ml). 2-8°C
9. Stop Solution (1×): A 0.5 N hydrochloric acid solution to stop the chromogen substrate reaction (11 ml). 2-8°C
Storage
1. Upon arrival, immediately store components of the kit at recommended temperatures up to the expiration date.
2. Store SP Conjugate and Biotinylated Antibody at -20°C.
3. Store Standard, Microplate, Diluent Concentrate (10×), Wash Buffer, Stop Solution, and Chromogen Substrate at 2-8°C.
4. Unused microplate wells may be returned to the foil pouch with the desiccant packs and resealed. May be stored for up to 30 days in a vacuum desiccator.
Performance Characteristics
1. The minimum detectable dose of human Lp(a) as calculated by 2SD from the mean of a zero standard was established to be 0.78 ng/ml.
2. Intra-assay precision was determined by testing three plasma samples twenty times in one assay.
3. Inter-assay precision was determined by testing three plasma samples in twenty assays.

General Description
Lipoprotein(a) [Lp(a)] is an atherogenic lipoprotein particle formed by an assembly of LDL particles and Apo(a) bound to Apo-B100 component of LDL. Apo(a), the main constituent of Lp(a), has serine proteinase activity and is capable of autoproteolysis. Apo(a) has 4548 amino acids, variable sizes from 200 – 700 kDa, multiple isoforms, and structural homology with plasminogen. It competes with plasminogen for its binding site, inhibiting tissue-type plasminogen activator 1 and leading to reduced fibrinolysis. The mean Lp(a) protein level ranges from 78 – 175 μg/ml depending on populations and the risk threshold is 300 μg/ml. High levels of Lp(a) in the blood is a risk factor for myocardial infarction (MI), coronary heart disease (CHD), cerebrovascular disease (CVD), atherosclerosis, thrombosis, and stroke.
