Abstract
Microorganisms inhabiting saline environments are an interesting ecological model for the study of the adaptation of organisms to extreme living conditions and constitute a precious resource of enzymes and bioproducts for biotechnological applications. We analyzed the microbial communities in nine ponds with increasing salt concentrations (salinity range 4.9-36.0%) of the Saltern of Margherita di Savoia (Italy), the largest thalassohaline saltern in Europe. A deep-metabarcoding NGS procedure addressing separately the V5-V6 and V3-V4 hypervariable regions of the 16S rRNA gene ofBacteriaandArchaea, respectively, and a CARD-FISH (catalyzed reporter deposition fluorescence in situ hybridization) analysis allowed us to profile the dynamics of microbial populations at the different salt concentrations. Both the domains were detected throughout the saltern, even if the low relative abundance ofArchaeain the three ponds with the lowest salinities prevented the construction of the relative amplicon libraries. The highest cell counts were recorded at 14.5% salinity forBacteriaand at 24.1% salinity forArchaea. WhileBacteriashowed the greatest number of genera in the first ponds (salinity range 4.9-14.5%), archaeal genera were more numerous in the last ponds of the saltern (salinity 24.1-36.0%). Among prokaryotes,Salinibacterwas the genus with the maximum abundance (similar to 49% at 34.6% salinity). Other genera detected at high abundance were the archaealHaloquadratum(similar to 43% at 36.0% salinity) andNatronomonas(similar to 18% at 13.1% salinity) and the bacterial "CandidatusAquiluna" (similar to 19% at 14.5% salinity). Interestingly, "CandidatusAquiluna" had not been identified before in thalassohaline waters.

Abstract
A new sample of tiragalloite from the Scerscen Glacier of the Malenco valley (Lombardy, Italy) has been studied by single-crystal X-ray diffraction and microchemical analyses. Structure refinement in space group P2(1)/n converge to R-1=0.0373 for 6149 reflections with I >= 2 sigma(I) and 0.0583 for all 8242 data. The refined unit-cell parameters are: a=6.6702(2) angstrom, b=19.9336(7) angstrom, c=7.5759(2) angstrom, beta=95.518(1)degrees, V=1002.63(5) angstrom(3); and the crystal-chemical formula is (Mn3.862+Ca0.10)(Sigma 3.96)(As0.855+V0.025+Si0.19)(Sigma 1.06) Si3O12(OH), with Z=4. The new data of tiragalloite from Scerscen confirm the general organization of the crystal structure, as previously reported in literature for two samples from Graveglia valley, Liguria, Italy, which contained a different amount of V5+ or As5+ cations. However, refinement of the site populations at the Mn(3) and Mn(4) sites suggests that the distribution of Ca among these atom sites might be different than that reported in literature. In particular, in tiragalloite from Scerscen, Ca seems to be preferentially located at the Mn-[6](4)(O5OH) octahedron rather than within the large Mn-[7](3)O-7 polyhedron. The topology of the [AsSi3O12(OH)](8-) unit of tiragalloite is compared with natural and synthetic phases containing similar groups of four tetrahedra.
