General Notes
This product (Catalog No. DPABH-26899) is a rabbit-derived polyclonal antibody raised against human TEX35. This antibody exhibits cross-reactivity with TEX35 from rat. It has been affinity purified by Protein A. EX35 is a protein-coding gene in humans, officially named testis expressed 35, that encodes multiple isoforms of the TEX35 protein, a testis-specific component associated with the microtubule cytoskeleton. Located on the long arm of chromosome 1 at position 1q25.2 (GRCh38: NC_000001.11, 178513109..178523510), the gene spans approximately 10.4 kb and consists of 11 exons, producing a primary transcript that translates into a 202-amino-acid protein containing a conserved Tsc35 domain characteristic of testis-specific proteins. First identified in 2006 as a novel testis-specific gene conserved between humans and mice, TEX35 exhibits restricted expression primarily in testicular tissues, with RNA levels reaching 26.3 RPKM in adult testis, and lower detection in certain fetal tissues such as adrenal gland, heart, intestine, kidney, lung, and stomach during 10-20 weeks gestation. The TEX35 protein localizes to the microtubule cytoskeleton, suggesting potential roles in cellular structure and dynamics within reproductive cells, though its precise molecular functions remain under investigation. Proteomic and interactome studies have implicated TEX35 in protein-protein interactions, including associations with the BRCA1-binding protein 2 (a negative regulator of nuclear import), the von Hippel-Lindau tumor suppressor pVHL in testicular contexts, and components of the human protein methyltransferase network, indicating possible involvement in pathways related to protein modification, ubiquitination, and spermatogenesis. Despite these associations, no direct links to human diseases or genetic variants have been established in major databases like ClinVar, and functional disruptions in model organisms have not yet revealed overt phenotypes. Orthologs of TEX35 are present in 85 species, including mice (Tex35, MGI:1920685), underscoring its evolutionary conservation, particularly in reproductive biology. Overall, TEX35 represents a specialized genetic element contributing to testicular cellular architecture, with ongoing research exploring its contributions to fertility and cytoskeletal regulation.
