This product (Catalog No. DPABH-22526) is a rabbit-derived polyclonal antibody raised against human PPOX, targeting the amino acid region 31–300 of the protein. This antibody exhibits cross-reactivity with Human PPOX from mouse and rat. It has been affinity purified by Protein A. Protoporphyrinogen oxidase (PPOX), encoded by the PPOX gene in humans (chromosome 1q22), is a flavin adenine dinucleotide (FAD)-dependent enzyme classified under EC 1.3.3.4 that catalyzes the oxygen-dependent, six-electron oxidation of the colorless precursor protoporphyrinogen IX to the red-colored protoporphyrin IX in the penultimate step of the heme biosynthetic pathway. This reaction involves sequential hydride abstractions and tautomerizations at the C10 methylene bridge, aromatizing the tetrapyrrole macrocycle while generating hydrogen peroxide as a byproduct, and is crucial for preventing non-enzymatic accumulation of reactive intermediates that could produce toxic reactive oxygen species. In aerobic organisms, PPOX ensures the controlled formation of protoporphyrin IX, which serves as the immediate precursor for heme insertion of iron by ferrochelatase to produce heme, a vital cofactor in hemoproteins like hemoglobin, myoglobin, and cytochromes essential for oxygen transport, respiration, and electron transfer.
DPABH-22526 is suitable for use in Western blot (WB) (1:500–1:1,000), IHC (1:20–1:200) applications.
DPABH-22526 is supplied as a liquid formulation in PBS with 0.02% sodium azide and 50% glycerol, pH 7.3. For long-term storage, keep at −20 °C. For short-term use, store at 2–8 °C. Avoid repeated freeze–thaw cycles to maintain antibody integrity and performance.
