This product (Catalog No. DPABH-04249) is a rabbit-derived polyclonal antibody raised against human PNP, targeting the amino acid region 1–289 of the protein. This antibody exhibits cross-reactivity with Human PNP from mouse and rat. It has been affinity purified by Protein A. Purine nucleoside phosphorylase (PNP; EC 2.4.2.1) is a ubiquitous enzyme found in eukaryotes and prokaryotes that catalyzes the reversible phosphorolysis of purine nucleosides, converting them to their corresponding purine bases and ribose-1-phosphate (or deoxyribose-1-phosphate for deoxy substrates), thereby facilitating the salvage and degradation of purines in nucleotide metabolism. This reaction proceeds via an S_N1 mechanism involving oxocarbenium ion formation at the ribose C1', with phosphate acting as a nucleophile stabilized by key residues such as His86 and Glu89 in the human enzyme. PNPs are classified into two main families based on oligomeric state and substrate specificity: low-molecular-mass homotrimers (∼90 kDa), like those in mammals, which prefer 6-oxopurines (e.g., inosine, guanosine), and high-molecular-mass homohexamers in many bacteria, which accommodate both 6-oxopurines and 6-aminopurines (e.g., adenosine).
DPABH-04249 is suitable for use in ELISA (1:4,000–1:8,000), IHC (1:50–1:100), Western blot (WB) (1:1,000–1:4,000) applications.
DPABH-04249 is supplied as a liquid formulation in 0.01M PBS, pH 7.4, 50% Glycerol, 0.05% Proclin 300. For long-term storage, keep at −20 °C. For short-term use, store at 2–8 °C. Avoid repeated freeze–thaw cycles to maintain antibody integrity and performance.
