The pyruvate dehydrogenase (PDH) complex is anuclear-encoded mitochondrial multienzyme complex that catalyzesthe overall conversion of pyruvate to acetyl-CoA and CO. Itprovides the primary link between glycolysis and the tricarboxylicacid (TCA) cycle, and thus is one of the major enzymes responsiblefor the regulation of glucose metabolism. The enzymatic activity ofPDH is regulated by a phosphorylation/dephosphorylation cycle, andphosphorylation results in inactivation of PDH. The protein encodedby this gene is one of the three pyruvate dehydrogenase kinasesthat inhibits the PDH complex by phosphorylation of the E1 alphasubunit. This gene is predominantly expressed in the heart andskeletal muscles. Alternatively spliced transcript variantsencoding different isoforms have been found for this gene.
