General Notes
This product (Catalog No. DPABH-01986) is a rabbit-derived polyclonal antibody raised against human IST1, targeting the amino acid region 1–186 of the protein. It has been affinity purified by Protein A. KIAA0174 is a human gene located on chromosome 16q22.2 that encodes the IST1 protein, a key component of the endosomal sorting complexes required for transport-III (ESCRT-III) machinery. The IST1 protein, consisting of 364 to 366 amino acids, features an N-terminal ELYC helical domain, a central proline-rich linker, and two C-terminal MIT-interacting motifs (MIMs) that facilitate binding to ESCRT-related proteins such as CHMP1A, CHMP1B, VPS4A, and VPS4B. Expressed ubiquitously across human tissues with highest levels in the kidney, IST1 is essential for the terminal stages of cytokinesis, where it recruits VPS4 ATPases to the midbody to sever intercellular membrane bridges, ensuring proper cell division; its depletion leads to abscission failure, resulting in multinucleated cells. Notably, IST1 interacts with spartin (SPG20) during cytokinesis and is not required for endosomal sorting or HIV-1 virus budding, distinguishing its role from other ESCRT components. Originally identified through cDNA sequencing from a human myeloid leukemia cell line, the gene—also known by synonyms CHMP8 and IST1—has no reported associations with human diseases in current genetic databases.
DPABH-01986 is suitable for use in ELISA (1:4,000–1:8,000), IHC (1:50–1:100), Western blot (WB) (1:1,000–1:4,000) applications.
DPABH-01986 is supplied as a liquid formulation in 0.01M PBS, pH 7.4, 50% Glycerol, 0.05% Proclin 300. For long-term storage, keep at −20 °C. For short-term use, store at 2–8 °C. Avoid repeated freeze–thaw cycles to maintain antibody integrity and performance.
